My research interest focuses on organelle biogenesis, in particular the manufacture of energy-transducing membranes in mitochondria and chloroplasts. Currently, the research going on in my laboratory can be divided into two main projects: 1) Elaboration of the redox pathways operating in the mitochondrial intermembrane space and the thylkoid lumen (funded by MDA and AHA) 2) Molecular dissection of mitochondrial complex I assembly (funded my UMDF). The first project is derived from my post-doctoral studies while the second one is a recent addition of my research investigation. I am also involved in a collaborative project with Dr. Meier on coiled-coil proteins. The goal is to define the function of a novel class of putative organellar coiled-coil proteins that was recently recognized. This project was funded by an OBPC grant to Dr. Meier where I am listed as a co-PI. I am also collaborating with Dr. Ghirardi (National Renewable Energy Laboratory) on increasing hydrogen production in the green alga Chlamydomonas reinhardtii. This project is funded by DOE.
Funding:I have pursued an aggressive strategy to secure extramural funding in order to support my research effort. Due to the large scope of my research, I have had multiple options for submission of my grant applications. I have secured a Muscular Dystrophy Association (MDA) research grant and my post-doctoral researcher Vincent Corvest was awarded an American Heart Association (AHA) post-doctoral fellowship. Both funding sources support on-going research focusing on aspects of mitochondria biogenesis. A UMDF grant was also awarded to me to support the research efforts on mitochondrial complex I. This is the first time that this medical agency funds plant-related research and this is the first grant from this agency to Ohio State University. In addition, both research projects benefit from a France-US grant where I am listed as a co-PI. This grant aims to support international collaboration at major american institutions for projects that are already on-going in the american host laboratory. The funds can only be used to support the visit of members from a french laboratory to the PI?s group. In addition, my collaborative work on hydrogen production with M. Ghirardi (NREL) was funded via a DOE subaward. This subaward is used to support the efforts of a graduate student working on this project in my laboratory.
Research: 1) The discovery that the thylakoid lumen, long assumed to be vacant, houses disulfide bonded proteins, suggests that thiol-disulfide chemistry is a catalyzed process in this compartment. Our recent findings indicate that distinct trans-thylakoid redox pathways operate in thio-reduction and thio-oxidation of luminal targets that still remain unidentified. We have identified LDR1 (Luminal Disulfide Reductase), a thioredoxin-like protein and LTO1 (Luminal Thiol Oxidase), a novel catalyst with disulfide bond formation activity as components of the reducing and oxidizing pathways, respectively. LDR1 is required for assembly of c-type cytochromes, a class of molecules that require reduction of a disulfide at a heme binding motif prior to the heme ligation reaction. The implication of LTO1 in plastid biogenesis and its relevant target of actions are currently being explored. In mitochondria, Cyc2p, a novel flavoprotein was identified as a redox assembly factor for c-type cytochrome. In vitro assays were developed in order to decipher the biochemical activity of the protein in the pathway and preliminary funding suggest a role as a NADPH-dependent apocytochrome c disulfide reductase for Cyc2p. Our contributions are very significant because thiol-based chemistry in the thylakoid lumen and the mitochondrial intermembrane space is a recent development in the field of membrane biogenesis. One manuscript describing some of our preliminary findings has been published.
2) A collaborative research project was started with Dr. Remacle (U. Liege, Belgium) in order to identify nuclear genes that control the assembly of complex I, a mitochondrial multimeric enzyme that is implicated in most of the mitochondrial disease in humans. It is believed that mutations in yet-to-be discovered complex I assembly genes are responsible for 50% of complex I-linked diseases that have no molecular explanation. We have developed the green alga Chlamydomonas as a genetically tractable system to isolate mutants that display a specific complex I assembly defect because complete loss of complex I is viable in this organism. Four amc mutants displaying a specific complex I defect (amc1 to amc4 for assembly of mitochondrial complex I) were isolated via several mutageneses. A candidate AMC2 gene coding for a novel mitochondrial-localized kinase has been identified and is now being tested. One manuscript describing our preliminary findings is published and one is under revision (Eukaryotic Cell). Our investigation in the green alga model indicate that the potential for novel discoveries in the field of complex I biogenesis is very high.
Funding: I will continue my effort toward securing extramural funding. The chloroplast aspect of my work will be targeted to NSF while the mitochondrial aspect of the work will be directed towards NIH. My conversation with my NSF program director makes me hopeful that this project is likely to be funded in the future. That the mitochondrial aspect of my research was funded by medical agencies such as AHA and UMDF makes me also confident that mitochondria-related research proposals can be targeted to NIH in the future.
Research: I will continue to develop a high quality, innovative and productive program with focus on the energy-transducing membrane systems in organelles and bring my publication output up to full speed. Furthermore, I will continue to promote the visibility of my work through invited presentations at research institutions and international conferences.
